The first nearly-complete skeleton was not found until 1966. It was missing only a foot and a tail. Many fossils are now kept at the Australian Museum.

There were three different species of Thylacoleo. The oldest was Thylacoleo hilli from the Pliocene. It was found in a cave in South Australia. Another species was Thylacoleo crassidentatus. It lived around 5 million years ago in Queensland. It was about the size of a large dog. The most famous was Thylacoleo carnifex. This species was the largest of them all. It reached weights of about 130 kg. This makes it similar in size to a modern lioness.

Thylacoleo was built for both climbing and fighting. It had very strong arms to grab its prey. Each hand had a large, retractable hooked claw. These claws could pull back into the hand. Some individuals may have been even larger. While scientists use dental remains to estimate size, these numbers are often considered dubious. Instead, researchers prefer using the circumference of limb bones for better accuracy.

The hunting mechanism of Thylacoleo was highly specialized. It possessed blade-like third premolar teeth called carnassials. These teeth were positioned much further forward in the jaw than in other mammals. It also had large first incisors that acted like canine teeth. While it had true canines, they were stubby and not very sharp. Pound for pound, Thylacoleo carnifex had the strongest bite of any known mammal. A 100 kg Thylacoleo had a bite force comparable to a 250 kg African lion.

This powerful bite allowed for a very efficient kill. Unlike modern big cats, Thylacoleo could not use a long, suffocating bite. Instead, it used its incisors to pierce flesh. Then, its specialized carnassials would crush the windpipe or sever major blood vessels. This included the carotid artery and the jugular vein. While an African lion might take 15 minutes to kill large prey, Thylacoleo could do it in less than a minute. However, this specialized skull was inefficient for catching small animals. This limitation may have contributed to its eventual extinction.

Thylacoleo was built for both power and movement. It had highly mobile and powerful forelimbs used to grapple with prey. Each hand had a single, large, retractable hooked claw. These claws sat on large, semi-opposable thumbs. This anatomy suggests they could deal a lethal blow. The animal also had climbing capabilities. Its hind feet had four functional toes. The first digit was reduced but had a roughened pad similar to a possum. This likely helped with climbing trees.

The anatomy of the tail also played a role in its lifestyle. Thylacoleo had specialized tail bones called chevrons. These bones strengthened the tail significantly. This strength likely allowed the animal to prop itself up on its hind legs. Such a position would be useful for climbing or attacking prey. The lumbar region, or lower back, was relatively rigid and straight. This suggests that the lower back was quite inflexible. These physical traits helped the genus occupy its unique ecological niche.

These ranged from small, possum-like animals to large, leopard-sized predators.
