Henderson was a master of the by-election, which is a special election held to fill a vacancy. He was elected to Parliament through five different by-elections in various locations. This allowed him to return to political life even after losing general elections.

Henderson reached the highest levels of the British government during times of crisis. In 1915, he became the first Labour member of the Cabinet. He served as President of the Board of Education. During the First World War, he was a member of the War Cabinet. In 1924, he served as Home Secretary in the first-ever Labour government. Later, in 1929, he became the Foreign Secretary to help improve relations with the Soviet Union.

His time as Foreign Secretary was marked by efforts to maintain global stability. He supported the League of Nations, an international organization meant to prevent conflict. However, the Great Depression caused a massive political crisis in 1931. The government was divided over whether to cut unemployment benefits by 10 percent. When Prime Minister Ramsay MacDonald formed a coalition with other parties, Henderson stayed with the main Labour Party. He led the party into a difficult election in 1931 where they won only 52 seats.

In his final years, Henderson focused entirely on preventing another world war.

He chaired the Geneva Disarmament Conference to reduce the number of weapons held by nations. His dedication to reducing military tension earned him the Nobel Peace Prize in 1934.
