In many species, such as birds, reptiles, and insects, the yolk is a specialized storage organ. This organ is constructed within the mother's reproductive tract. In smaller animals, like certain fish and invertebrates, the yolk material is not in a separate organ. Instead, the yolk sits directly inside the egg cell. The yolk is not a living cell like protoplasm. It is largely passive material called deutoplasm. These nutrients and control structures are supplied during a process called oogenesis, which is the formation of the egg.

The specific shade can change based on the hen's diet. For example, eating alfalfa can make yolks more yellow, while capsicum peppers can make them red.

Sometimes, eggs appear unusual due to biological variations. Double-yolk eggs occur when ovulation happens too quickly or two yolks join together. These are rare, and the chicks often struggle to hatch because they interfere with each other. Conversely, yolkless eggs are sometimes called "wind" eggs. These often happen when a young hen's reproductive system is not yet fully ready. In these cases, a piece of tissue might be wrapped in albumen and a shell, but it lacks the yolk needed for a chick to grow.

Beyond nutrition, the yolk has many practical uses. In cooking, it acts as an emulsifier for sauces like mayonnaise, custard, and hollandaise. It is also used in traditional art as a component of egg-tempera paint. In science, egg yolk agar is used to test for the presence of bacteria like Clostridium perfringens. The yolk even contains an antibody called antiglobulin, or IgY. This antibody is transferred from the hen to the yolk through passive immunity. This helps protect the embryo and the hatchling from invading microorganisms.
