He was appointed to this role on 8 October 2010 by Prime Minister Vladimir Putin. On 9 August 2011, he was chosen to lead the Russian delegation for the 2012 London Summer Olympics. He later became the Russian Deputy Minister of Sports on 18 June 2012. These roles required him to manage large groups of athletes and officials.
His career in sports administration included a significant and difficult period involving the World Anti-Doping Agency, known as WADA. WADA is an international organization that works to ensure sports remain fair and clean. Kolobkov served as Russia's representative to this agency. However, he faced intense scrutiny regarding laboratory data. WADA found that Russia was falsifying doping data, which refers to the illegal use of substances to improve performance. Kolobkov unsuccessfully denied these findings. He was criticized by top Russian athletes for potentially endangering their professional careers.
In late 2015, the situation with WADA reached a critical point. On 18 November 2015, WADA declared the Russian Anti-Doping Agency to be non-compliant. This means the agency was not following the required international rules. As a result, Kolobkov lost his position as a WADA representative in January 2016. He was also barred from serving on the organization's Foundation Board. This period of controversy led to Russia being barred from the Olympics and other international competitions for four years. British lawyer Jonathan Taylor, who wrote a report on these sanctions, questioned Kolobkov's competence or integrity. 
Despite these challenges, Kolobkov continued to hold high office in the Russian government. On 11 August 2016, he was appointed a Class 2 State Advisor of the Russian Federation. Shortly after, on 19 October 2016, he became the Russian Minister of Sport. He held this powerful position until 15 January 2020. At that time, President Putin dismissed him from the role and replaced him. 
Throughout his life, Kolobkov has received numerous prestigious honors for his contributions. In 1992, he was named a Merited Master of Sports. He was also awarded the Order of Honour on 19 April 2001.
