He later moved to Santo Domingo to fight Spanish forces. During this time, he met the famous commander Toussaint Louverture. Dessalines eventually became a chief lieutenant to Louverture. By 1799, he had reached the rank of brigadier general.

The conflict changed when the French government underwent shifts. Napoleon Bonaparte rose to power in France. Napoleon wanted to restore slavery in Saint-Domingue to protect sugar profits. In 1802, a French expeditionary force arrived under General Charles Leclerc. Dessalines fought the French at the Battle of Crête-à-Pierrot. During this battle, his 1,300 men defended a fort against 18,000 attackers. They held the position for 20 days before retreating due to shortages.

After Louverture was captured and died in a French prison, Dessalines took command. He became the leader of the revolution on 18 May 1803. He led his forces to a decisive victory at the Battle of Vertières. This battle took place on 18 November 1803. His forces defeated the French army led by Donatien de Rochambeau. This victory allowed the French to surrender their last territory on 4 December 1803.

Haiti was declared an independent republic on 1 January 1804. Dessalines was chosen by a council of generals to lead the nation. He oversaw the 1804 massacre of the remaining French population. This event killed between 3,000 and 5,000 people. Some historians suggest he did this to prevent a French reinvasion. He also granted citizenship to certain groups, such as German and Polish soldiers. However, tensions remained with the mixed-race population.

Dessalines was later named Emperor of Haiti, ruling from 1804 to 1806. He married Marie-Claire Heureuse Félicité Bonheur, who became empress. Together, they had or adopted 16 children. Dessalines used autocratic rule to manage the nation's economy. He enforced plantation labor to help the country grow. His reign ended in 1806 when he was assassinated by members of his own administration.
