His uncle, George Clinton, was a very famous leader too. DeWitt worked as his uncle's secretary before starting his own career. He joined the Democratic-Republican Party to help lead the state. He used his skills to win many different elections.

He held several high-ranking offices, including United States senator and mayor of New York City. Most notably, he served two terms as the governor of New York. Clinton was a key figure in the Democratic-Republican Party during the early 1800s. His political career was defined by his belief in infrastructure. Infrastructure refers to the basic systems, like roads or canals, that a society uses to function. He believed these projects could drive economic growth and encourage people to participate in politics.

Clinton's rise to power began through family connections and early political appointments. He was the nephew of George Clinton, a two-term U.S. vice president and governor of New York. DeWitt served as his uncle's secretary before entering the New York State Legislature in 1798. In 1801, his uncle appointed him to lead the Council of Appointments. This council held the power to appoint thousands of public officials, such as sheriffs and mayors. Because the council was loyal to the Clinton family, DeWitt became a very powerful politician. He used this influence to manage patronage, which is the power to give jobs to supporters.

His early political life included intense rivalries and significant challenges. In 1802, while serving as a director of the Manhattan Company, he clashed with supporters of Aaron Burr. This conflict led to a duel in New Jersey with John Swartwout. After this, Clinton served in the U.S. Senate from 1802 to 1803. He resigned from the Senate because he was unhappy with living conditions in Washington, D.C. He then moved back to New York to serve as mayor. He held the mayoralty for several terms between 1803 and 1815.

Clinton also participated in national politics during a major election cycle. In 1812, he ran for the presidency of the United States. He challenged the incumbent president, James Madison. Clinton ran with support from the Federalist Party and some antiwar Democratic-Republicans. Although he lost the election, he received 89 electoral votes. This was a strong performance for a Federalist-supported candidate. He carried most of the Northeastern United States during this campaign.

As governor, Clinton focused his energy on the construction of the Erie Canal.

He was inspired by Jesse Hawley to support a canal connecting Lake Erie to the Hudson River. Many critics at the time doubted the project's success. They mockingly called it "Clinton's Folly" or "DeWitt's Ditch." Despite this, Clinton secured $7 million from the legislature in 1817 to begin construction. This amount was a massive investment for the time. He oversaw the project through his various terms as governor.

The Erie Canal was completed in 1825 and became an immense success.

To celebrate, Clinton traveled on the packet boat Seneca Chief. He even poured water from Lake Erie into New York Harbor to symbolize the connection of the East and West. The canal revolutionized how goods were moved. The cost to move freight between Buffalo and Albany dropped from $100 to $10 per ton. This efficiency allowed the state to recoup its money through tolls. The canal's success also changed how the public viewed Clinton.

Beyond politics, Clinton was active in many social and scientific organizations. He helped organize the New-York Historical Society in 1804 and served as its president. He also led the American Academy of the Fine Arts. Clinton was a high-ranking Freemason and served as Grand Master of the Grand Lodge of New York. In 1826, he dealt with the William Morgan Affair, a scandal involving the disappearance of a man. Clinton issued proclamations offering rewards for information regarding the case. He remained a dedicated leader in New York until his death in 1828.
